Abstract

Essence

In situ sequencing mapped base and prime editing directly in mouse and macaque tissues and showed RNA-LNP liver editing across metabolic zones.

Evidence

This in vivo platform study validated imaging-based ISS in edited mouse brains and applied it to mouse and macaque livers after adenine base editor RNA-LNP dosing, including repeated-dose experiments.

Caveat

It measured spatial editing rates and dosing distribution in animal tissues, not therapeutic correction, safety, or disease outcomes.
