A detection limit of 1.24 pM for microRNA-21 was achieved using a newly engineered steric-regulation framework for Cas12a.

  • The framework allows for predictable tuning of Cas12a activation through engineered extensions on split activators.
  • Systematic analysis identified rules governing the assembly and activation of Cas12a based on extension orientation and length.
  • The incorporation of sterically regulated split activators into a DNA circuit enhanced coordination and reduced background leakage.
