Sensitive detection of Ec-16S rDNA using 3D DNA rollers and CRISPR/Cas12a with amplified light signals

Updated

Abstract

The ECL biosensor demonstrates a detection limit of 27.29 aM for Ec-16S rDNA.

  • A novel ECL biosensor was developed using CRISPR/Cas12a technology combined with a cascade amplification strategy.
  • The cascade amplification strategy includes strand displacement amplification and dual-particle three-dimensional DNA rollers.
  • The biosensor achieved exponential amplification of the target signal, significantly enhancing sensitivity.
  • Manganese dioxide nanoflowers were used to increase the ECL intensity of the biosensor.
  • The biosensor operates effectively within a response range of 100 aM to 10 nM.
