Using Mass Spectrometry to Reveal How Semaglutide Molecules Change Shape and Group Together

Updated

Abstract

Semaglutide forms oligomers up to 10-mers within 24 hours under specific conditions.

  • Low micromolar concentrations of Cu2+ and Zn2+ ions may enhance the formation of higher-order and more abundant oligomers.
  • The formation of oligomeric species is associated with hydrophobic interactions at the C-termini in a parallel arrangement.
  • N-termini and lipid chains are loosely engaged, allowing for high-affinity binding to human serum albumin (HSA).
  • Glycation of HSA may further facilitate the dissociation of oligomeric species.
