Short-term oral safety of salcaprozate sodium (SNAC) in two common rat types

Abstract

At a dose of 2000 mg/kg/d, 50% of female SD rats treated with Salcaprozate sodium (SNAC) showed mortality over 13 weeks.

  • No mortality was observed in Wistar rats at doses up to 1000 mg/kg/d over 13 weeks.
  • Slightly altered electrolyte levels and lower globulin levels were noted in both SD and Wistar rats, though these remained within historical control ranges.
  • Increased liver and kidney weights were found in SNAC-treated rats, potentially indicating an adaptive response without histopathological changes.
  • Histopathological changes in the stomach were likely due to irritation from the dosing method.
  • A no-observed-adverse-effect level (NOAEL) for SNAC in SD rats could not be determined, while the NOAEL for Wistar rats was identified as 1000 mg/kg/d.
