Improved under-the-tongue delivery of a modified diabetes drug leads to long-lasting effects in diabetic mice

Abstract

A 3 mg/mL TE-8105 liquid formulation at pH 7.6 with a 5-minute mucosal contact yielded optimal glycemic control.

  • Fatty acid chains may enhance mucosal permeability for peptide delivery.
  • Phenol (0.3-0.5%) and mannitol (4%) improved efficacy when combined, exceeding their individual effects.
  • TE-8105 demonstrated similar therapeutic potency (∼7%) to semaglutide via the subcutaneous route.
  • TE-8105's prolonged plasma half-life allows dosing every two days, compared to daily administration for semaglutide.
  • The findings support the development of a needle-free sublingual delivery system for GLP-1 therapies.
