Reducing early inflammation after radiation may lessen long-term bone damage

Abstract

An early increase in pro-inflammatory SASP proteins was observed following radiotherapy in prostate cancer patients.

  • Cellular senescence is linked to radiotherapy-related bone loss.
  • Clearance of senescent cells may alleviate radiation-associated bone deterioration.
  • Ruxolitinib, a Janus kinase inhibitor, was effective in reducing radiation-related bone loss in preclinical studies.
  • Acute elevation of SASP markers was confirmed in both human patients and mouse models following irradiation.
  • Early suppression of SASP using JAK inhibitors reduced signs of chronic bone deterioration and cellular dysfunction.
