Lipid Nanoparticles Using Molecular Recognition Boost Cancer Immunotherapy by Changing the Tumor Environment

Updated

Abstract

Lipid nanoparticles (LNPs) modified with a sequential pretargeting strategy improve intratumoral retention and cellular uptake.

  • The use of adamantane-conjugated antibodies primes tumors for enhanced capture of cucurbit[7]uril-functionalized LNPs.
  • Incorporation of NLG919 into the LNPs may inhibit IDO-mediated tryptophan breakdown, reducing regulatory T cell activity.
  • The delivery platform allows encapsulation of circular RNA that encodes an IL-2-Fc fusion protein to stimulate intratumoral cytokine production.
  • This approach may result in improved antitumor immune responses, leading to reduced tumor growth and prolonged survival.
