Highly sensitive molecular detection using paired DNA-cutting enzymes activated together by a special DNA hairpin amplifier

Updated

Abstract

A detection limit of 55 aM was achieved using a novel dimeric palindromic hairpin probe for molecular diagnostics.

  • The dimeric palindromic hairpin probe integrates multiple functions, including target recognition and signal amplification, into a single structure.
  • Cyclic amplification processes enable the production of palindromic trigger strands that activate two Cas12a complexes simultaneously.
  • Quantitative detection ranges from 1 fM to 1 nM were observed, indicating high sensitivity.
  • The assay demonstrated high sequence specificity and differentiated fluorescence responses in miRNA extracts from healthy individuals and breast cancer patients.
  • This approach reduces design complexity and instability compared to traditional systems requiring multiple probes.
