A mouse brain tumor model mimics key molecular, immune, and functional traits of human glioblastoma to support treatment research

Updated

Abstract

Murine glioblastoma models with patient-specific mutations exhibit striking resemblance to human disease.

  • The models developed diffuse, infiltrative lesions similar to those observed in human glioblastoma.
  • Tumors displayed a tumor immune microenvironment primarily composed of microglia with minimal lymphocyte presence.
  • Intrinsic resistance to immune checkpoint blockade may stem from poor antigen presentation and T cell priming.
  • mEGFRvIII-PC cells showed significant resistance to antigen-specific T cell-induced cytotoxicity and promoted T cell exhaustion.
  • Stimulation with murine interferon-gamma enhanced PD-L1 upregulation compared to MHC-I levels.
