For KCNJ13 L144P retinal disease models, synonymous or bystander edits undermined base editing while prime editing restored Kir7.1 function.

Evidence

Patient variant analysis plus human iPSC-derived RPE and HEK293 cell editing experiments compared base and prime editing effects on L144P Kir7.1 channel activity.

Caveat

Functional rescue was shown in engineered cell models, with about 20% prime editing and restored channel activity in 28% of edited cells (12/43), not in animal or human outcomes.
