Selective gene editing in the cyanobacterium Cyanothece PCC 7425 using CRISPR/Cas12a

CRISPR/Cas12a editing enables the rapid generation of knockout mutants in Cyanothece sp. PCC 7425.

  • Four genes related to metabolism and stress response were successfully deleted using CRISPR/Cas12a.
  • Full chromosome segregation was achieved after just one round of selection for all mutants.
  • Genotypic and phenotypic characterization confirmed the successful knockout of targeted genes.
  • The sodB mutant demonstrated targeted deletion through biochemical analysis.
  • This system establishes Cyanothece sp. PCC 7425 as a model for studying oxidative stress pathways and biomineralization.
