Abstract

An antibody-based targeting bispecific-LNP platform enables targeted delivery of lipid nanoparticles to specific cell types.

  • TbsAb-LNP complexes can simultaneously bind to LNP polyethylene glycol motifs and specific cell surface markers.
  • The platform allows for increased accumulation of mRNA-LNPs in targeted cell types through colocalization.
  • Endosomal efficiency kinetics of TbsAb-LNPs were assessed using various bispecific loading ratios.
  • Targeted delivery was successfully demonstrated in vitro with xhuCD3-expressing lymphoblasts and xmTfR1-expressing cell lines.
  • In vivo targeting was achieved in xmTfR1-expressing cells, indicating potential for broader applications.
  • The technology may enhance clinical vaccine and T-cell delivery strategies by enriching TbsAb-LNPs to splenic cells.
