A commercial mRNA-lipid nanoparticle sample (approx. 4200 b) was used to refine a customizable analysis method.

  • Optimal conditions for mRNA integrity analysis included sample precipitation by isopropanol and high urea concentrations.
  • The absence of formamide as a sample diluent improved assay sensitivity and resolution.
  • High concentrations of dye were associated with enhanced sensitivity in the method.
  • Methodological challenges were identified in assessing mRNA integrity due to interactions between mRNA and the sample matrix.
