Liver organoids formed on a thermoresponsive substrate exhibited higher levels of liver-specific gene expression and tissue functions compared to traditional cell-laden constructs.

  • Mesenchymal stem cells and a thermoresponsive substrate facilitated the formation and retrieval of liver organoids.
  • Organoids self-organized into viable sheets that detached from the substrate when cooled.
  • The organoid-laden constructs showed enhanced protein synthesis, ammonia detoxification, and drug metabolism.
  • Histological analysis indicated a 'histomimetic' cellular organization and expression of tissue-specific markers in organoid-laden constructs.
  • The MSC-laden substrate may serve as an effective platform for creating intact liver organoids suitable for 3D bioprinting.
