Blocking Tim3 and A2a receptors changes the cancer-fighting ability of engineered immune cells in a human cervical tumor model

Abstract

Knockdown of A2aR alone or with Tim3 improved CAR T cell function against solid tumors.

  • Inhibition of A2aR and Tim3 enhanced proliferation, cytokine production, and cytotoxicity of CAR T cells in the presence of tumor cells.
  • Both double knockdown CAR T cells and control CAR T cells effectively controlled tumor growth in a human cervical cancer model.
  • Single knockdown CAR T cells were associated with reduced survival in mice.
  • Concomitant targeting of Tim3 and A2aR may improve CAR T cell therapy efficacy in solid tumors, but caution is warranted.
