Effects of GLP-1R activators on hunger and fullness neurons vary with time and metabolism in living animals

Abstract

Liraglutide and semaglutide increase activity in proopiomelanocortin (POMC) neurons and inhibit Neuropeptide Y/Agouti-related peptide (NPY/AgRP) neurons in a time-dependent manner.

  • The activation of POMC neurons by liraglutide and semaglutide is dependent on GLP-1 receptors and requires TRPC5 subunits.
  • An increase in excitatory input to POMC neurons from glutamatergic cells also depends on TRPC5 subunits.
  • Liraglutide and semaglutide decrease excitatory input to NPY/AgRP neurons through the activation of K-ATP and TRPC5 channels in GABAergic neurons.
  • The effects on POMC and NPY/AgRP neurons are influenced by the nutritional state of the animals, varying between fed and food-deprived conditions.
