Daily changes in liver gene activity and metabolism are altered in MASLD

Abstract

A cohort of 290 patients with liver biopsies reveals significant alterations in gene expression and metabolite patterns associated with metabolic dysfunction-related liver disease.

  • Time-of-day-dependent gene expression patterns in human liver are significantly altered in steatotic and metabolic dysfunction-associated steatohepatitis (MASH) cases.
  • Core molecular clock components remain robust in both steatotic and MASH livers, indicating some preservation of circadian rhythms.
  • Hundreds of transcripts involved in cell communication, signaling, and metabolism show stage-specific alterations based on the time of day.
  • Rhythmic patterns of amino acids and lipids in the metabolome are disrupted in pathological liver conditions.
  • Key signaling pathways, including TNFα and PPARγ, are predicted to contribute to the observed changes in rhythmicity.
