How Adding Sugar to Carotenoid Breakdown Products in Different Leaf Tissues Helps Maintain Carotenoid Balance in Arabidopsis

Abstract

Overexpression of phytoene synthase (PSY) in Arabidopsis led to a dramatic accumulation of β-carotene in roots and nongreen calli.

  • Enhanced pathway flux from PSY overexpression resulted in increased phytoene synthesis rates, but carotenoid levels in leaves remained unchanged.
  • Surplus PSY in chloroplasts was partially soluble and inactive, with the active portion localized in the membrane.
  • Increased pathway flux did not lead to higher long-chain apocarotenals but resulted in elevated levels of C13 apocarotenoid glycosides (AGs).
  • The majority of AGs formed were identified as being primarily produced by CCD4, a carotenoid cleavage dioxygenase.
  • Tissue-specific capacities to synthesize xanthophylls influenced the modes of carotenoid accumulation and apocarotenoid formation.
