Abstract

CTCF-dependent boundary integrity at the NOTCH1 locus is associated with transcriptional regulation.

  • NOTCH1 is located within a defined topologically associated domain with CTCF/cohesin occupancy at both ends.
  • Deletion of boundary CTCF sites in human K562 cells increased transcription of NOTCH1 and associated non-coding transcripts.
  • Disruption of boundary elements impaired cell proliferation and clonogenic growth.
  • Chromatin profiling indicated defects in domain insulation and changes in regulatory interactions within the NOTCH1 domain.
  • Cross-species analysis revealed that while domain architecture is conserved in mice, the effects of boundary disruption varied by cell type.
