Using targeted nanoparticles to deliver precise DNA editing for lung cancer treatment

Updated

Abstract

A targeted TALE base editing lipid nanoparticle may effectively block KRAS expression in lung cancer models.

  • Non-small cell lung cancer (NSCLC) accounts for approximately 85% of lung cancer cases.
  • The KRAS oncogene mutation is significant in the development and spread of NSCLC.
  • A transcription activator-like effector (TALE) base editor was developed to convert a C base to a T base in the KRAS gene.
  • This conversion generates a stop codon that can inhibit KRAS expression.
  • The targeted TALE lipid nanoparticle demonstrated base editing capabilities and cancer targeting in mice with NSCLC.
  • Therapy efficacy was observed in NSCLC cancer-bearing mice treated with the targeted TALE LNP.
