Gene activity linked to seed dormancy and its loss in a hormone-insensitive mutant of Arabidopsis

Abstract

The germination of the sleepy1-2 (sly1-2) mutant can reach 74% with overexpression of the GA receptor GID1b.

  • Increased seed dormancy in sly1-2 is linked to the accumulation of DELLA proteins, which repress germination.
  • After-ripening for 11 months resulted in 51% germination, demonstrating a potential pathway to alleviate dormancy.
  • Half of the transcriptome changes during after-ripening depend on the SLY1 gene's role in DELLA protein degradation.
  • Overexpression of GID1b activated changes in only 23 genes, indicating that significant germination changes can occur with minimal gene expression alterations.
  • GID1b overexpression appears to drive early germination processes, followed by potential negative feedback regulation later in imbibition.
