How tRNA Needs Change in Different Tumor Environments

Updated

Abstract

An atlas of tRNA dependencies was established through large-scale CRISPRi perturbation mapping across 16 cancer cell lines.

  • tRNA dependencies during tumor growth exhibit tissue organization at isodecoder resolution.
  • These dependencies are associated with codon demand at the isoacceptor level.
  • In vitro culturing reorganizes tRNA dependencies, highlighting environmental sensitivity.
  • Protein components like aminoacyl-tRNA synthetases showed largely preserved dependencies across environments.
  • A shared response across cancer cell lines indicates a common axis of isoacceptor dependency remodeling.
  • Specific tRNA isoacceptors are linked to adaptation to the tumor environment and differential translation related to proliferation.
