A highly sensitive CRISPR-based sensor for detecting toxin-producing Clostridioides difficile

Abstract

The developed biosensor achieves a detection limit of pM level for C. difficile DNA within 40 minutes.

  • A novel crRNA was engineered to specifically recognize the toxin B gene (tcdB) of C. difficile.
  • Activation of Cas12a's activity occurs upon binding to the target, leading to the cleavage of ssDNA reporters.
  • The biosensor demonstrated high specificity against non-target pathogens.
  • Robust stability of the biosensor was observed over 7 days.
