A universal safety switch for CRISPR/Cas12a that works without chemicals or outside triggers

Updated

Abstract

Essence

A programmable RNA inhibitor could switch activated CRISPR/Cas12a complexes back toward inactivity without chemical modification or external stimulation.

Evidence

Platform experiment using experiments and simulations tested iRNA strand-displacement control of Cas12a ribonucleoproteins, including autonomous intracellular suppression of uncontrolled editing while preserving PCSK9 knockout.

Caveat

The abstract reports validated Cas12a experiments and cellular editing control, but not therapeutic efficacy, organism-level safety, or clinical outcomes.
