Maximum mutation load caused by a CRISPR-Cas9 gene drive element

Updated

Abstract

Essence

In yeast, CRISPR-Cas9 gene-drive elements appeared to add little or no genome-wide mutational burden.

Evidence

A mutation-accumulation experiment in Saccharomyces cerevisiae measured de novo mutation and loss-of-heterozygosity rates with a CCGD or Cas9 alone.

Caveat

Power calculations only bound any rate effect to less than about 30%, and weak LOH length or distribution signals need testing outside yeast.
