How lipid nanoparticle treatment that disrupts blood vessels affects the tumor environment

Updated

Abstract

The combination therapy achieved robust vascular disruption and antitumor efficacy in CT26 and Renca tumors in BALB/c mice.

  • Efficacy of the combination therapy was significantly lower in B16-F10 and LLC tumors in C57BL/6 mice.
  • In refractory LLC tumors, gene silencing efficiency in tumor endothelial cells was approximately 40%, compared to around 79% in responsive tumors.
  • Enhanced extracellular matrix remodeling and increased innate immune activation in the tumor microenvironment may contribute to greater susceptibility to therapy.
  • Higher expression of the IFNAR1 receptor, which mediates type I interferon signaling, was found in responsive tumors.
