WenDan Decoction significantly improved depressive-like behaviors in high-fat diet mice while showing relatively low hepatorenal toxicity.

  • WenDan Decoction restored intestinal mucosal integrity and mitigated inflammation in high-fat diet mice.
  • The treatment re-established gut microbial balance and regulated the metabolism of arachidonic acid.
  • A strong negative correlation was found between the gut bacteria Lactobacillus murinus and the arachidonic acid metabolite PGF2α.
  • WenDan Decoction reduced blood-brain barrier disruption and decreased pro-inflammatory metabolites.
  • The treatment inhibited the PI3K/AKT/mTOR pathway and restored autophagic function, improving neuronal health.
  • Fecal microbiota transplantation confirmed the roles of gut microbiota and PGF2α in the antidepressant effects of WenDan Decoction.
