Abstract

This method enables the detection of SARS-CoV-2 mutations within 1 hour, achieving a detection limit of 7.49 aM.

  • The biosensor detects both single- and double-stranded amplification products of SARS-CoV-2 mutant variants.
  • It generates double-stranded DNA products with PAM sequences and single-stranded DNA products without PAM sequences.
  • Both types of amplification products can be recognized by the crRNA, enhancing detection sensitivity.
  • The assay shows 100% sensitivity and 100% specificity in detecting SARS-CoV-2 mutations in 106 clinical samples compared to DNA sequencing.
  • High specificity is observed in differentiating mutant variants from wild-type sequences.
