Fast test using CRISPR for detecting Babesia parasites in ticks and livestock blood

Abstract

Reported cases of human babesiosis in the United States increased from 1,742 in 2014 to 3,586 in 2023.

  • Cattle babesiosis is associated with significant economic losses in livestock production, estimated at hundreds of millions of US dollars annually.
  • A one-pot assay was developed for genus-level detection of Babesia spp., minimizing contamination risks compared to conventional methods.
  • The assay can detect Babesia spp. within 40 minutes at 37°C, targeting a conserved region of the 18S rRNA gene.
  • High analytical sensitivity was demonstrated, with a limit of detection of 5 copies/µL for recombinant plasmid templates.
  • Field validation of the assay showed positive rates of 15.49% in pooled tick samples and 9.43% in clinical blood samples from cattle and sheep.
  • The assay displayed 100% concordance with conventional PCR methods for both specimen types.
