Genetic and pharmacologic alterations of actin structure and function are associated with reduced lifespan and physiological defects.

  • Knockdown of actin and actin-binding proteins led to premature disruption of filament organization.
  • ABP knockdowns induced a transcriptome profile resembling that of aged organisms.
  • Actin dysfunction exacerbated age-related issues, including mitochondrial dysfunction and impaired autophagy.
  • Pharmacological destabilization of actin mirrored genetic knockdowns, while mild stabilization modestly extended lifespan.
  • Analysis of human genome data indicated correlations between common gene variations and age-related gait speed decline.
