Viral tools for RNA-targeting Cas13d: Comparing high-accuracy versions DjCas13d and hfCas13d

Updated

Abstract

High-fidelity Cas13d (hfCas13d) consistently caused significantly fewer transcriptome alterations compared to DjCas13d when targeting highly expressed genes.

  • Both hfCas13d and DjCas13d demonstrate similar knockdown potential for various RNA transcripts.
  • DjCas13d occasionally outperformed hfCas13d in knockdown efficiency.
  • hfCas13d is associated with a reduced risk of cleaving unintended bystander RNAs compared to DjCas13d.
  • The effects on transcriptome alterations were evaluated using RNA sequencing and other methods.
