Special Microfluidic Devices with Wing-Like Shapes for Faster Testing and Scaling of Lipid Nanoparticle Production

Abstract

A high-throughput microfluidic platform enables the continuous production of lipid nanoparticles (LNPs) at flow rates up to 50 mL min.

  • The platform utilizes unique micromixer designs to enhance mixing and LNP formation across a broad range of flow rates (0.2-50 mL min).
  • Minimal reagent volumes as low as 0.1 mL are required for up to liter-scale production.
  • Simultaneous synthesis of multiple LNP formulations is achieved with sizes ranging from 38-150 nm and a polydispersity index (PDI) of less than 0.2.
  • Performance validation includes mRNA and pDNA transfection, as well as siRNA uptake studies in various cell lines (HEK293, A549, CFBE).
