Astragaloside IV may protect kidney filtering cells in diabetic kidney disease by restoring cell recycling and preventing cell death through blocking Notch1 signaling via SIRT6 activity

Updated

Abstract

AS-IV treatment restored nephrin and podocin expression while reducing glomerular basement membrane thickness (p < 0.01) in diabetic nephropathy mice.

  • AS-IV has shown efficacy in reducing proteinuria and renal tubular damage in diabetic nephropathy models.
  • It activates the deacetylase SIRT6, which modifies histones and suppresses Notch1 expression.
  • This suppression may enhance autophagy and decrease apoptosis, aiding in the repair of renal filtration barrier injury.
  • AS-IV treatment reversed high glucose-induced podocyte apoptosis and autophagy dysfunction (p < 0.01).
  • Direct binding of AS-IV to SIRT6 was confirmed, with increased SIRT6 occupancy at the Notch1 promoter observed.
