Berberine improves liver cell insulin resistance by controlling daily rhythm genes that regulate sugar and fat metabolism

Abstract

Treatment with 20 μmol·L~(-1) berberine improved glucose consumption and reduced lipid levels in insulin-resistant HepG2 cells.

  • Berberine treatment increased the expression and nuclear localization of BMAL1 and CLOCK in insulin-resistant HepG2 cells.
  • Levels of adiponectin, insulin receptor substrate 2, and phosphatidylinositol 3-kinase regulatory subunit p85 were up-regulated by berberine.
  • Phosphorylated Akt and mTOR levels were also increased following berberine treatment.
  • Conversely, berberine down-regulated levels of certain proteins associated with lipid synthesis, such as ChREBP and fatty acid synthase.
  • The addition of CLK8 decreased glucose consumption and altered the levels of key metabolic regulators in these cells.
