BLA inputs are the dominant excitatory pathway onto D1 medium spiny neurons in the nucleus accumbens core.

  • The nucleus accumbens integrates cognitive, emotional, and motor functions but its circuitry is not fully understood.
  • A hierarchy of synaptic inputs was observed based on the type of medium spiny neuron (MSN) targeted.
  • D1 MSNs receive stronger inputs from the basolateral amygdala (BLA) compared to the prefrontal cortex (PFC) and ventral hippocampus (vHipp).
  • In contrast, PFC inputs are more dominant on D2 MSNs, followed by vHipp and BLA inputs.
  • Endocannabinoids contribute to pathway- and cell-specific synaptic plasticity, affecting excitatory circuits in a differentiated manner.
  • CB1 receptors uniformly depress excitatory pathways regardless of MSN type, while TRPV1 receptors control inputs in a pathway-specific way.
