Disrupted body clocks in colorectal cancer and the potential of timed treatment as personalized medicine

Circadian disruption is associated with colorectal cancer (CRC) through multiple mechanisms.

  • Deregulated expressions of core clock genes disrupt essential cellular processes such as cell cycle control and DNA repair.
  • Circadian disruption is linked to key CRC signaling pathways including Wnt/β-catenin, PI3K/AKT/mTOR, and Notch signaling.
  • The altered intestinal clock can lead to changes in gut microbial distribution and impaired intestinal barrier function.
  • Chronological misalignment may contribute to both intratumoral and intertumoral heterogeneity in CRC tumors.
  • Deregulation of clock genes is recognized as a potential prognostic biomarker for CRC and is associated with treatment resistance.
  • Optimizing chemotherapy and radiotherapy based on individual biological clocks may improve efficacy and reduce toxicity in advanced CRC patients.
