Highly sensitive and multiple detection of microcystin-LR using a CRISPR-based three-mode sensor

Abstract

Detection of microcystin-LR (MC-LR) was achieved with a limit as low as 1.0 aM using a novel tri-mode aptasensor.

  • The sensor employs an aptamer-based competitive displacement reaction for specific recognition of MC-LR.
  • Activation of Cas12a's trans-cleavage activity occurs in the presence of MC-LR, enabling multiple signal outputs.
  • Fluorescence detection is performed using a digital droplet fluorescence system that enhances local fluorescence signals.
  • The detection range for the sensor extends from 1.0 × 10^-13 M to 1.0 × 10^-6 M.
  • Analytical performance in real samples showed recoveries between 81.26% and 113.21% (n = 3).
