Rapid and ultra-sensitive detection of COVID-19 protein using a surface-bound CRISPR sensor with enhanced fluorescence

Abstract

The platform achieves a limit of detection at 67.2 fg/mL within 30 minutes for SARS-CoV-2 nucleocapsid protein.

  • A one-pot biosensor integrates metal-enhanced fluorescence and nanoscale spatial confinement for rapid protein detection.
  • The biosensor features an ultrabright fluorescence intensity more than 860-fold greater than that of free auramine O.
  • Upon binding the target protein, the system can produce a signal reduction of up to ∼85% from the initial signal.
  • The signal-to-noise ratio of the biosensor is 83.89, which is approximately 2.5 times higher than that of a solution-phase system.
  • The platform demonstrates good sensitivity, selectivity, stability, and recovery in bronchoalveolar lavage fluid.
