Fluorescent and visual tests using CRISPR to detect and identify different groups of mpox virus

Updated

Abstract

The dual-mode detection platform achieved 100% sensitivity for Clade II detection using fluorescence methods.

  • A novel dual-mode detection platform was developed to distinguish between Clade I and Clade II of the mpox virus.
  • The platform integrates two technologies: recombinase-aided amplification and CRISPR-Cas12a, enabling both laboratory and field use.
  • Detection sensitivities were reported as 1 copy/reaction for the universal assay and 10 copies for the Clade I-discriminatory assay using fluorescence.
  • Both assays demonstrated high specificity, successfully differentiating mpox from other related viruses.
  • Clinical validation indicated 100% sensitivity and 80% specificity for the universal assay compared to quantitative PCR.
  • Further evaluation with authentic Clade I clinical specimens is needed to confirm the platform's typing performance.
