Abstract

Essence

HEAL, a compact inducible dCas12f CRISPRa system, enabled strong endogenous gene activation and showed therapeutic-like effects in mouse models.

Evidence

A CRISPRa platform-engineering study tested HEAL in vitro and in vivo, reporting over 100,000-fold gene activation plus AAV interleukin-10 activation in acute kidney injury mice and ChemHEAL TSLP activation in obese mice.

Caveat

These are preclinical platform and mouse-disease demonstrations, so they do not establish human gene-therapy efficacy or delivery safety.
