Changes to RNA patterns improve prime editing in living cells and boost different editing methods

Abstract

A single injection of lipid nanoparticles achieved nearly 70% editing efficiency in the bulk mouse liver.

  • Engineered prime editing guide RNAs (pegRNAs) with modified RNA motifs were developed to improve editing efficiency.
  • The approach resulted in a near 80-fold increase in editing efficiency compared to conventional pegRNAs.
  • Incorporating densely modified RNA motifs was broadly applicable across various RNA sequences and editing platforms.
  • The method demonstrated up to an 11-fold increase in base editing efficiency.
  • Systemic delivery of the prime editor in RNA format could enhance the therapeutic potential of prime editing.
