Risk of Depression with GLP-1 Diabetes Drugs Compared to Other Diabetes Medicines in Adults with Type 2 Diabetes

Abstract

Nine studies involving 1,735,325 patients found no significant difference in the risk of developing depression between GLP-1 RA users and other antidiabetic agents (HR 1.05; 95% CI 0.97-1.14).

  • Sensitivity analysis indicated that excluding one influential study resulted in a significant hazard ratio of 1.09 (95% CI 1.01-1.19; p = 0.035).
  • Substantial heterogeneity was observed among the studies (I² = 83.2%).
  • Limited evidence suggested potential benefits in anxiety-related outcomes, although these findings were heterogeneous.
  • Reported adverse events were mainly gastrointestinal in nature.
  • Overall, GLP-1 RA use does not appear to be linked to an increased risk of depression in adults with type 2 diabetes.
