How DNA methylation blockers affect changes in the body’s internal clock caused by light

Abstract

Application of the DNA methyltransferase inhibitor RG108 near the suprachiasmatic nucleus significantly blocks light-induced period after-effects.

  • Period after-effects on circadian behavior from changing light conditions are disrupted by RG108.
  • Single light pulses that normally reset circadian rhythms have their period after-effects significantly reduced by RG108.
  • RG108 also prevents period after-effects on clock gene rhythms following phase resetting induced by vasoactive intestinal peptide.
  • Another DNA methyltransferase inhibitor, SGI-1027, blocks period after-effects from optogenetic stimulation of the SCN.
  • Acute clock resetting does not seem to depend on DNA methylation, unlike subsequent changes in circadian period.
