Using stabilizing mutations to improve RSV pre-fusion F mRNA vaccine design

Abstract

The TriM-5 mRNA vaccine candidate induced 3.03-fold higher neutralizing antibody titers against RSV A2 compared to the Moderna RSV mRNA sequence.

  • Codon optimization of the TriM-5 mRNA resulted in approximately threefold increases in IgG antibody titers.
  • The TriM-5 mRNA-LNP showed significantly higher secretion levels of immune markers IFN-γ, IL-5, and IL-17A.
  • Both TriM-5 mRNA-LNP and Moderna mRNA-LNP effectively reduced lung viral loads in virus challenge experiments.
  • No obvious lung pathology was observed in mice immunized with the TriM-5 mRNA-LNP.
