Electrochemical sensor detecting tuberculosis DNA using CRISPR and a redox-labeled DNA probe

Abstract

The biosensor can detect the tuberculosis biomarker IS6110 at a limit of detection of 14.5 nM within 60 minutes.

  • The biosensor utilizes the trans cleavage activity of CRISPR-Cas12a to identify target DNA associated with tuberculosis.
  • Detection is achieved through the interaction of target DNA with a non-specific ssDNA substrate, Poly T.
  • Various concentrations of target DNA were tested, confirming the biosensor's effectiveness in detecting TB-related biomarkers.
  • This platform represents the first reported use of a CRISPR-Cas12a-based electrochemical sensor specifically for tuberculosis.
