New CRISPR-based sensor for detecting circulating tumor DNA

Abstract

The ECL-RET biosensor demonstrates a detection limit as low as 3.0 fM for circulating tumor DNA (ctDNA).

  • The biosensor utilizes Ru(phen) as an energy donor embedded in double-stranded DNA and Au nanocages as the energy acceptor.
  • It is based on the target-activated CRISPR-Cas12a system, which enhances the efficiency of resonance energy transfer.
  • The detection range of the biosensor spans from 10 fM to 1 nM for ctDNA.
  • The system effectively recognizes the L858R mutation in ctDNA from non-small cell lung cancer patients.
