Designed CRISPR-Cas9 Tool for Improving Bacterial Transfer into the Fungus Phanerochaete chrysosporium

Abstract

An optimized Agrobacterium-mediated transformation protocol was developed for Phanerochaete chrysosporium, achieving stable integration of T-DNA.

  • Transformation efficiency was enhanced by low-temperature incubation at 23°C and the addition of acetosyringone.
  • Stable expression of the green fluorescent protein (sGFP) was confirmed in transformed mycelia.
  • The modified CRISPR-Cas9 compatible vector demonstrated potential for stable maintenance in P. chrysosporium.
  • Although gene editing was not achieved, the findings support future applications in metabolic engineering.
  • This work establishes a foundation for advancing lignin valorization and sustainable biochemicals production.
