Designed mRNA nanoparticles allow controlled protein production and immune activation for new treatments

Updated

Abstract

Self-assembling mRNA origami (mRNA-OG) can influence protein output and immune response.

  • mRNA-OG forms compact, programmable RNA nanostructures that encode protein-coding sequences.
  • Folded mRNA-OG recruits ribosomes similarly to unfolded constructs but results in lower protein production.
  • The compact structure of mRNA-OG improves encapsulation by cationic lipid delivery systems.
  • Different RNA architectures elicit distinct cytokine responses in human dendritic cells.
  • Folded mRNA-OG induces a modest integrated stress response, which is not fully explained by standard signaling pathways.
