Improved Transformer-Based Tool for More Efficient DNA Editing

Abstract

The engineered transformer base editor (etBE) achieved up to 35.13% efficient base editing in a humanized mouse model.

  • etBE shows a significant editing efficiency improvement of up to 35.11-fold compared to the parental transformer base editor (tBE).
  • High editing fidelity is maintained while keeping off-target mutations at low background levels.
  • In vivo application targeting the PCSK9 gene resulted in a 24% reduction in plasma PCSK9 protein levels.
  • Low-density lipoprotein cholesterol (LDL-C) levels decreased by 33% following etBE treatment.
  • Minimal off-target mutations were observed in the process.
